Detection of the Latent Functional Insufficiency of Dopaminergic Neurons in the Nigrostriatal System in a Chronic Model of Parkinson’s Diseaseстатья

Аннотация:The dopaminergic nigrostriatal system is a key regulator of motor function in human beings. The degradation of the system is an important component of pathogenesis of Parkinson's disease. The disturbances of motor function as well as the opportunity of diagnosis appear 20-30 years after the beginning of the pathological process of dopaminergic neurons death (50-60% of cells) and decrease of dopamine synthesis level in striatum (70-80% of dopamine). Currently, the absence of effective therapy is associated with the late diagnosis of the disease when there are almost no targets for pharmacotherapy. Therefore, the priority of modern neurology is the development of preclinical diagnosis of Parkinson's disease. In this work a new approach of dopaminergic nigrostriatal system functional deficiency assessment with dopamine synthesis inhibitor (alpha-methyl-p-tyrosine) treatment was developed on experimental chronic models of preclinical Parkinson's disease on mice. The administration of alpha-methyl-p-tyrosine leads to presence of motor dysfunction in experimental group, but not in the group of control.
The data obtained show the possibility of using of this approach as a provocation test for the preclinical diagnosis of Parkinson's disease, which allows starting a preventive treatment aimed to slow down the death of dopaminergic neurons.
