Аннотация:Tissue-specific promoters play a crucial role in bothfundamental research and practical applications. Theyallow targeted gene expression only in specific celltypes, minimizing off-target effects on neighboringcells. In the context of gene therapy for vasculardiseases, the use of tissue-specific promoters becomesparticularly important to minimize non-targetedtransgene expression and potential side effects.Endothelial cells, which line blood vessels and regulatethe exchange of substances between blood and organs,are potential targets for gene therapy in certainvascular diseases.Several endothelial-specific promoters have beenproposed and characterized to date, including Tie1 andTie2, PECAM-1, ICAM-1, E- and P-selectins, Flt1 andflk-1, endoglin, eNOS, claudin 5, VE-Cadherin,preproendothelin-1 and von Willebrand factor. In thisreview, we discuss the characteristics and applicationsof these endothelial-specific promoters. We highlighttheir advantages and limitations and explore theirpotential in gene therapy.
