Sex-dependent protective effect of combined application of solubilized ubiquinol and selenium on the development of monocrotaline(mct)-induced pulmonary hypertension in Wistar ratsстатьяТезисы

Аннотация:Selenium (Se) is part of a large number of first-line antiperoxidant defense enzymesin the body. The positive effect of the CoQ10 and Se combination was demonstratedagainst the background of CVD treatment, so the study of this combination in models of PH associated with inflammation, such as monocrotaline (MCT)-induced PH,is of great interest.Methods: The study was performed according to a unified protocol on male (♂) andfemale (♀) Wistar rats (180-210g) and followed the Council Directive 86/609/EECprinciples. On the day 1 rats of 4 groups received a single injection of 60 mg/kg ofMCT water-alcohol solution sub/c to reproduce the model of PH; the control groupsreceived only an equivalent volume of the water-alcohol solution (♀ were injectedduring diestrus). On days 7 and 14 the MCT groups were injected into the tail veinwith 1.solubilized U (MCT-U group) (30 mg/kg) or 2. only a vehicle for U substance(MCT-V), or 3. Were given selenium (Solgar, 100 𝜇g) 10 𝜇g/kg per os. (Se-MCT) andthen daily, or 4. together Se daily and U (MCT-Se-U); 5.Control ♂ and ♀ groupswere also intravenously injected with vehicle (Control). On day 22 in anesthetizedrats (urethane 1.2 g/kg) we measured right ventricle systolic pressure (RVSP) andthe degree of RVH (RV/body weight*1000) to assess the development of PH.Results: 3 weeks after the MCT injection an increase of RVSP relative to control wasshown in all MCT-groups of ♂ and ♀ by an average of 10±5%, except MCT-Se-Ugroup of ♂ (Control 38.5±5.5 mmHg vs MCT-Se-U 43.5±3.1 mmHg, p>0.05).Increase in RV mass was found only in the MCT-V and MCT-Se groups by anaverage of 26±4% in ♂ and 21±2% in ♀. Lung mass (g) in the MCT-U groups of♂ and ♀ did not differ significantly from the Control groups (♂Control 1.45±0.24 vsMCT-U 1.78±0.40 MCT-Se-U 1.53±0.31, p>0.05; ♀Control 1.45±0.27 vs MCT-U1.57±0.23, MCT-Se-U 1.68±0.11, p>0.05), while it was significantly higher in theMCT-V and MCT-Se groups. The use of Se and U combination revealed a protectiveeffect on lung mass only in ♂, but not in ♀.Conclusions: Double U administration has a protective effect against the development of 3-week MCT-PH by reducing H and lung mass in animals of both sexes.Combination of U with Se also has a protective effect that was demonstrated bythe reduction of RVH in ♂ and ♀, but the reduction of RVSP and lung weight is shownonly in ♂-rats. We conclude that combined U and Se are more effective in preventionPH development in ♂ than in ♀.
