Thymoquinone Induces Mitochondrial Damage and Death of Cerebellar Granule Neuronsстатья

Аннотация:Thymoquinone (TQ) exhibits a wide spectrum of biological activities. Most research on the neurotoxic action of TQ is carried out on the models of cancer cell lines. The present study was designed to test the TQ toxic action in primary neuronal cultures in vitro. Here we show that the application of 0.04–0.05 mM TQ for 24 h induced a high dose-dependent death rate of cultured cerebellar granule neurons (CGNs). Using a CellROX Green and MitoSox Red, the cell death was shown to be preceded by an increase in the radical’s production. Confocal and electron microscopy demonstrated that 0.05 mM TQ for 5h induced unusual change in the localization of mitochondria in cell, an increase in the size of these organelles and swelling of cells. Antioxidant N-acetyl-L-cysteine (NAC, 2 mM) significantly protected CGNs from this toxic action.
Taken together, these facts lead us to suggest that TQ can be toxic for normal neurons and ROS-dependent processes of mitochondrial destruction can be one of the major causes of neuronal damage and TQ-induced death.
