Аннотация:The effect of a phase transition of a thermosensitive matrix on the temperature dependence of equilibrium and kinetic constants of individual steps of enzymatic reactions was studied using alkaline phosphatase and alpha-chymotrypsin immobilized in poly-N-isopropylacrylamide gel as a model system. The phase transition of the thermosensitive gel matrix was shown to affect weakly the temperature dependence of equilibrium parameters of the enzymatic reactions. All the reversible effects of enzyme activity reduction with increasing temperature are exclusively due to the temperature dependence of kinetic constants for individual steps of the enzymatic process.
