Plant stress-tolerance is a central challenge in modern intensive agriculture, and an area of intense fundamental research. The photosynthetic apparatus (PSA) is a key target for protective mechanisms, with non-photochemical quenching of chlorophyll excited states in the antenna being one of the most important. In higher plants and their closest evolutionary relatives, char algae, induction of non-photochemical quenching is achieved due to the thylakoid membrane protein PsbS, a component of photosystem I. In other divisions of the algae, LhcSR, a protein similar to those of the photosynthetic antenna, performs this function (it also works in char algae and mosses). It is assumed that the evolutionary replacement of LhcSR with PsbS is associated with the transition to terrestrial life. Several years ago, it was discovered that the expression of PsbS gene could be induced in the green alga Chlamydomonas reinhardtii in response to stress conditions for a short time (in an hour time scale). Consequently, it has been postulated that PsbS may serve as an ancillary protective mechanism in green algae during a brief transitional period at the onset of stress. Recently, we have observed prolonged (in a day timescale) expression of the PsbS gene in response to low positive temperature in psychrotolerant green microalgae Lobosphaera incisa IPPAS-C2047. These new data may lead to a revision of the role of PsbS in protecting PSA from excessive illumination in Chlorophytes, as well as the timepoint of its appearance in oxygenic phototrophs during evolution. The aim of this project is to identify conditions for long-term induction of the PsbS-coding gene expression in different orders of green algae, and to conclude whether this phenomenon is typical for Chlorophyta.
