Реакции гидроксилирования и Байера-Виллигера активно используются в фармацевтической промышленности и тонком органическом синтезе. Как правило, такие реакции проводятся с помощью ферментов - цитохром Р450 оксидоредуктаз (CYP450) и монооксигеназ (МО). Использование этих ферментов позволяет получить целевые продукты с выходом до 99-100% и в виде только одного оптического изомера с ее 98-99% и выше. Кроме органических субстратов для проведения обоих реакций необходимы кислород и восстановленная форма кофермента NADPH. Последний является очень дорогим соединением (более 10 000 Евро за кг) и для снижения стоимости процесса используется вторая ферментативная реакция – восстановление образующегося в основной реакции NADP+ обратно до NADPH. Наиболее эффективным вторым ферментом является мутантная NADP+-специфичная формиатдеги¬дро¬геназа (ФДГ). Для получения новых высокоэффективных вариантов NADP+-специфичных ФДГ были определены трехмерные структуры двух ферментов из бактерий и дрожжей и на основании анализа полученных структур проведены эксперименты методом направленного мутагенеза. Ферменты используют или в очищенной форме в виде пар CYP450-ФДГ и МО-ФДГ, или попарно ко-экспрессируют в клетках E.coli. Однако вследствие невысокой удельной активности CYP450 и МО общая производительность невысока. Для повышения эффективности процесса получены генетические конструкции, экспрессирующие гибридные белки, содержащие в одной полипептидной цепи последовательности ФДГ- CYP450, CYP450-ФДГ (по два варианта каждой) и ФДГ-МО (с двумя вариантами ФДГ) Все гибридные ферменты были получены в активной форме, выделены, очищены, охарактеризованы и протестированы в ряде реакций. Показано, что сближение активных центров двух ферментов в гибридном белке приводит к увеличению общей каталитической эффективности всего процесса в несколько раз.
