In the result of large-scale screening a significant antibiotic activity for cultural broth sample of bacterium Amycoloplasis sp. was shown. Following study kindly provided by our colleagues who detected Tetracenomycin X (ThX) as an active substance of cultural broth. This compound was mentioned in literature as a promising antibiotic, however, a mechanism of antibiotic activity has not previously been studied. According to data obtained using reporter strain E.coli pDualrep2 we supposed translational inhibition as a probable mechanism of antibiotic activity. This suggestion was proved by an ability of purified ThX to translational inhibition in vitro. Afterwards 32P toe-printing method demonstrated an inhibition of translation and formed a hypothesis that interruption of translation happens on an elongation stage. Therefore, an antibiotic activity of ThX due to inhibition of translation on elongation stage was determined. Thereby this compound might be a perspective molecule as a potential antibiotic and this process requires a further study.In future we are planning to study a mechanism of inhibition, to find out rRNA bases involved in this process and, finally, estimate if ribosome stopping is sequence-dependent or not.
