Due to dramatic increase of the resistance to wellknown antibiotics, discovery of new antibacterial agents becomes one of the principal goal of the modern biochemistry. For acceleration of drug discovery process, we created a simple, reliable and costeffective system for highthroughput identification of a potential antibiotic mechanism of action. Two fluorescent proteins with inducible expression were used for double reporter system, which allows not only antimicrobial activity detecting, but also for simultaneous sorting of potential antimicrobials into those that cause ribosome stalling, and others that induce SOS response due to DNA damage. The mechanism of action determination is necessary for rational improvement of the found compound; rapid identification of drug target allows significantly decrease time and coast of the molecule optimization process. By means of this system, more than 100 thousands of different synthetic and natural compounds were tested, and several new antibacterial agents were found.
