Аннотация:Cells of the Gram-negative obligate methylotrophic bacterium Methylobacillus flagellatum KT, which can only use methanol and methylamine as sole carbon and energy source, oxidize 1,2-propanediol to lactaldehyde and lactate by using a pyrroloquinoline quinone (PQQ) containing cytochrome c dependent methanol dehydrogenase and an NAD+-dependent aldehyde dehydrogenase, both of which have a broad substrate specificity. The kinetics of 1,2-propanediol oxidation and lactic acid appearance in the buffered medium were followed, and K(m) for substrate, pH, and temperature optima were determined. M. flagellatum KT cells immobilized in polyacrylamide, agarose, or carrageenan gels maintain the ability to oxidize 1,2-propanediol to lactic acid. The half-life of the process with the cells in carrageenan was increased 5-6 times compared with free cell suspensions.
